Leukocyte transendothelial migration（白细胞跨内皮迁移）

基础研究核心：阐明白细胞（中性粒细胞、单核细胞、淋巴细胞）从血管内穿越内皮细胞层进入炎症组织的多步骤分子机制，包括 “滚动 - 黏附 - 迁移” 三个关键阶段。首先，白细胞通过 Selectin 家族（L-Selectin、E-Selectin）与内皮细胞表面的配体结合，产生滚动效应；随后，白细胞表面的 Integrin（αLβ2、α4β1）被激活并与内皮细胞的 ICAM-1、VCAM-1 结合，实现稳定黏附；最后，白细胞通过 PECAM-1 介导的同型相互作用，穿越内皮细胞间隙（依赖 Rho GTPases 调控的细胞骨架重排）。研究重点包括 Integrin 激活的信号机制、内皮细胞间隙的动态调控、不同白细胞亚型的迁移差异，以及炎症性疾病（如关节炎、哮喘）中迁移过程的异常增强机制。
